I had the same problem happen to me.

Simple fix is to rename DfrgNtfs.exe (you need to take ownership to rename)
in C:\Windows\System32. This is a work around as now you can’t defrag your
hard drive if it is NTFS.

Other avenues I’m looking at is turning off scheduled defrag and only doing
manual. The problem seems to only crop up when the user is away from the
keyboard. If Defrag missed a scheduled defrag, it will run when the
computer is idle and this is when it causes the computer to lockup. I am
guessing here, but I think if you miss too many defrags, it gets queued up
by Task Scheduler and it keeps calling for defrag and defrag ends up getting
preempted by itself and gets stuck in a loop. I say this because the reason
I originally renamed DfrgNtfs.exe was it would start without user
intervention and start using up 25% CPU time on one of four cores (I have a
Q9550 Core 2 Quad) but hardly any hard drive activity, as much as you would
get when it is idling. So something about scheduled defrag is causing a
problem, especially if you missed a few.

DfrgNtfs when it starts in this condition blocks pretty much all reads or
writes to the hard drive. I can minimize windows, but I can’t close any
apps or open new ones, not even Task Manager. Opening or closing apps
requires reading or writing to the hard drive. I right-click on Desktop to
bring up properties and Desktop becomes greyed out and eventually I have to
hit the reset button. I tried the power button but it doesn’t shut down
because defrag is blocking I/O to the hard drive.

So I turned off defrag on schedule (dfrgui.exe) and disabled ScheduledDefrag
under Task Scheduler, it’s buried pretty far down. Go to Computer
Management -> System Tools -> Task Scheduler -> Microsoft -> Windows ->
Defrag and in the top middle pane, listed under ManualDefrag you find
ScheduleDefrag. Disabled it and changed the file name back to DfrgNtfs.exe
and running defrag manually.
